Can after the flight come back to the Mode "Preperaring for Bording"???

After unloading, have you tried clicking on a passenger load again? This seems to start the process over again.

I found out because I didn't spot the 'unload' option and wondered how to make the passengers disembark. I was stopped, parking brake on, engines off, door open, but they still wouldn't leave. So I tried clicking the 'medium' load and my plane was suddenly empty, with passengers then making their way inside. After that I spotted 'unload' lurking there to the right of the other options.

You might not see the "preparing for boarding" status text, but hopefully shouldn't stop you from making another flight. Worth a try, anyway :D

Yes, if you have unloaded and your flight has been recorded in the captain's log, you can start all over again by simply clicking any of the realtime load presets.
